Channels are the rooms of your voice server. Permissions decide what each person can do in each one. You need admin rights first: How to become admin on your Mumble server.
Create channels
Right-click the Root channel (or any channel) and choose Add.
Give it a name. Add a description if you want people to see what it is for.
Drag channels to reorder them, or drop one onto another to make it a sub-channel.
A simple layout for a gaming group:
Lobby (everyone starts here)
Game 1
Game 2
Raid team (private)
AFK
Staff
Where new people arrive
New users start in the Root channel, so name it something welcoming and keep a short description on it. To change the starting channel you edit the defaultchannel setting in murmur.ini (under Settings, Config files), using the channel's numeric ID. Most groups simply use Root as the lobby.
Understand ACL rules
On a channel choose Edit, then ACL. Each rule says: for this user or group, allow or deny a permission, in this channel and optionally its sub-channels. Rules are checked from the root channel down, and a rule lower down can override one above it.
Useful permissions:
Enter: go into the channel
Speak: talk in it
Whisper, Text message: private speech and chat
Move, Kick, Mute/Deafen: moderator tools
Make channel, Write: create and edit channels
A private channel
To make Raid team private:
Open Edit, Groups and create a group raid. Add the registered members.
Open ACL and add a rule for group all that denies Enter.
